CJC-1295 & Ipamorelin

CJC-1295 is understood to bind the growth hormone-releasing hormone receptor (GHRH-R) on pituitary somatotrophs, while Ipamorelin engages the growth hormone secretagogue receptor (GHS-R1a), the same receptor engaged by endogenous ghrelin. Occupying these two distinct receptor populations simultaneously is understood to produce complementary stimulation of the same target cell. Ipamorelin is relatively selective, with limited affinity for receptors that drive cortisol or prolactin release.

At the GHRH receptor, signaling proceeds through Gs-coupled activation of adenylyl cyclase, rising cyclic AMP, and downstream protein kinase A activity, which is linked to growth hormone gene transcription and somatotroph priming. The GHS-R1a pathway instead couples to Gq, activating phospholipase C and mobilizing intracellular calcium that may facilitate vesicular GH release. These two signaling arms are understood to converge in a way that supports a larger, more physiologic secretory pulse than either input alone.

Because GHS-R1a is also expressed in hypothalamic and other central regions, the blend's ghrelin-mimetic activity is understood to relate to appetite signaling and aspects of CNS regulation of the somatotropic axis. These central effects are generally framed around feeding and neuroendocrine modulation rather than direct cognitive or mood action. Beyond this, limited direct neural activity is described for this blend at the doses used for endocrine effects.
